Abstract

|
The serologic diagnoses which are faster and differential are an important preceeding step to control and treat infectious diseases. Therefore, development of a new method to diagnose diseases has been required continuously to improve more
sensitivity
and specificity at the level of higher technology.
We have tried to develop a new diagnostic method of hemorrhagic fever with renal syndrome (HFRS) due to Hantaan viral infection. For the rapid, simple and specific diagnosis of HFRS, we developed immunoblot analysis for the detection of
antibodies
against Hantaan virus by using biotin/streptavidin system.
Partially purified Hantaan nucleocapsid viral protein(NP) was prepared from the infected mice brain and separated by SDS-PAGE and transferred electrically to nitrocellulose. The transfeffed protein was reacted first with specific antibody
following
reaction with biotinylated secondary antibody. The immunocomplex was reacted with a streptavidin-alkaline phosphatase. The sharp band was observed by incubation with BCIP/NBT chromogenic substrates.
As compared to immunofluorescent antibody(IFA) test, this method was verified to be more sensitive and more specific to diagnose HFRS patients. Therfore, this result suggests the immunoblot analysis using biotin/streptavidin is an advanced method
for
rapid, specific and sensitive diagnosis of HFRS.
